(+)-Discodermolide is an architecturally intriguing marine sponge natural product possessing significant antitumor activity.  To explore the potential of (+)-discodermolide as a lead cancer chemotherapeutic agent, we, in collaboration with Kosan Biosciences, have developed a program aimed toward the following:

  • (1) developing detailed structure-activity relationships

  • (2) characterizing the (+)-discodermolide binding domain of tubulin

  • (3) design and synthesis of novel analogs whose tumor cell growth inhibitory activity is superior to that of the natural product.
